Dennis Slamon at UCLA identified HER2 amplification as an oncogene in breast cancer and championed the development of trastuzumab (Herceptin) against medical and industry skepticism. His persistence in developing trastuzumab for HER2-positive breast cancer transformed outcomes for the 20-25% of breast cancer patients with HER2 amplification — from among the worst prognosis to among the best when treated with HER2-targeted therapy. Slamon is a founding figure of modern precision oncology.
