Dennis Briand focuses on developing PROTAC degraders for historically undruggable oncoproteins, with particular emphasis on KRAS variants including KRAS G12C, extending the therapeutic reach of targeted protein degradation beyond what covalent inhibitors alone can achieve. His laboratory has addressed the oral bioavailability challenge of PROTACs through macrocyclization and prodrug strategies that reduce molecular weight while maintaining ternary complex formation. He has developed cell-permeable PROTAC probes for KRAS family members with different activating mutations. His work is advancing the next generation of KRAS degraders toward clinical candidacy.
