Sandra Demaria has elucidated the fundamental immunological mechanisms underlying the abscopal effect, discovering the critical role of the cGAS-STING cytosolic DNA sensing pathway in connecting radiation-induced DNA damage to type I interferon production and dendritic cell activation. Her mechanistic studies have revealed that specific radiation dose and fractionation schedules optimally activate anti-tumor immunity through distinct molecular pathways. She has established the scientific framework for rational design of radiation-immunotherapy combinations.
