Greg Delgoffe demonstrated that tumors outcompete T cells for glucose in the tumor microenvironment, creating metabolic insufficiency that impairs anti-tumor T cell effector function, and that PD-1 signaling exacerbates this metabolic competition by suppressing T cell glycolysis and mitochondrial biogenesis. His innovative approach of mitochondrial transfer to tumor-infiltrating lymphocytes restored their metabolic fitness and anti-tumor killing capacity, providing a novel strategy for metabolic rescue of exhausted T cells. He develops metabolically enhanced adoptive cell therapies including mitochondria-boosted CAR-T cells with superior persistence and function in metabolically hostile tumor microenvironments.
