G. David Roodman is a pioneer in osteoclast biology and bone metastasis who has made foundational contributions to understanding how cancer cells, particularly myeloma cells, corrupt the normal RANK/RANKL signaling pathway to drive osteoclastogenesis and bone destruction. His studies identified DKK1 as a key mediator of myeloma bone disease that suppresses osteoblast function while simultaneously stimulating osteoclasts. His laboratory work provided critical rationale for clinical development of denosumab and RANKL pathway inhibitors in multiple myeloma and solid tumor bone metastases.
