David Lyden discovered the pre-metastatic niche concept, demonstrating that primary tumors secrete exosomes and soluble factors that condition distant organs before cancer cells arrive, recruiting bone marrow-derived cells and remodeling the ECM to create a receptive microenvironment for metastatic colonization. His landmark work showed that tumor-derived exosome integrins determine organ-specific metastatic tropism, with exosome integrin alpha-6/beta-4 directing lung metastasis and integrin alpha-v/beta-5 directing liver metastasis. He identified VEGFR1-positive bone marrow progenitors as key architects of the pre-metastatic niche. His discoveries have created opportunities for intercepting metastasis before cancer cells colonize distant sites.
