David Lane co-discovered p53 in 1979 and subsequently dedicated his career to developing therapeutic strategies to reactivate p53 function in cancer. He established the concept of using MDM2 inhibitors to prevent p53 degradation in tumors retaining wild-type TP53. His pioneering work on small molecules and peptides that can restore wild-type conformation to mutant p53 proteins led to clinical development of APR-246 and related compounds. He has championed international p53 research through the p53 Family organization and collaborative networks.
