Darren Bhatt has advanced understanding of ABC transporter-mediated resistance to tyrosine kinase inhibitors, demonstrating that ABCG2 (BCRP) and ABCB1 (P-glycoprotein) actively efflux multiple TKIs including imatinib, gefitinib, and sunitinib, limiting their intracellular accumulation and therapeutic efficacy. His research established pharmacogenomic biomarkers in ABC transporter genes that predict TKI response variability across patients. He showed that newer-generation TKIs were designed to be poor ABC transporter substrates, explaining their improved efficacy over first-generation agents. His work on transporter-drug interactions has informed rational TKI design and dosing optimization strategies.
