Darrell Irvine is a leading engineer of nanoparticle vaccine delivery systems and saponin-based adjuvant platforms for cancer immunotherapy, having developed albumin-hitchhiking vaccines and self-assembling nanoparticles that dramatically improve antigen and adjuvant delivery to lymph nodes for potent anti-tumor immune priming. His work demonstrated that saponin-based adjuvants including ISCOM-like structures and Matrix-M formulations generate superior CD8+ T cell responses compared to conventional adjuvants through enhanced antigen cross-presentation. He has developed amphiphilic adjuvant conjugates that self-assemble into nanoparticles for lymph node targeting. His engineering approaches have produced some of the most potent cancer vaccine formulations in preclinical development.
