Daniel De Carvalho has established the paradigm of viral mimicry in cancer, demonstrating that DNA demethylating agents reactivate endogenous retroviruses to trigger innate immune signaling through dsRNA/MDA5/MAVS pathways. His work provided the scientific rationale for combining DNMT inhibitors with immune checkpoint blockade, now in clinical trials. He has shown that ERV-derived peptides serve as tumor-associated neoantigens recognized by T cells.
