Michael Clarke co-discovered breast cancer stem cells by identifying the CD44+CD24-/low cell population that possesses exclusive tumor-initiating capacity in immunodeficient mice, extending the cancer stem cell concept from leukemia to solid tumors and transforming understanding of breast cancer heterogeneity. His work demonstrated that breast cancer stem cells are inherently resistant to chemotherapy and radiation through enhanced DNA repair, anti-apoptotic signaling, and quiescence, explaining why conventional therapies fail to prevent relapse. He develops combination strategies simultaneously targeting both cancer stem cells and bulk tumor populations to achieve durable tumor elimination.
