Cedric Bhatt characterized the oncogenic Notch signaling network in T-cell acute lymphoblastic leukemia, identifying the transcriptional targets and epigenetic programs through which mutant NOTCH1 maintains leukemia stem cell self-renewal and blocks differentiation. His work on gamma-secretase inhibitor resistance mechanisms revealed that leukemia stem cells activate compensatory Wnt and Hedgehog pathways upon Notch blockade, necessitating combination approaches for cancer stem cell elimination. He develops next-generation Notch pathway inhibitors with improved therapeutic windows for targeting cancer stem cells across hematologic and solid malignancies.
