Carl Allen at Texas Children's has made contributions to understanding Langerhans cell histiocytosis (LCH) — a myeloid neoplasm with dendritic cell characteristics, now understood to be driven by MAPK pathway activation (BRAF V600E in ~50%). His work establishing LCH as a clonal myeloid neoplasm rather than an inflammatory disease, and on BRAF inhibitor activity in LCH, has transformed understanding of this disease.
