Carolyn Bertozzi, Nobel Laureate in Chemistry 2022, invented LYTAC (Lysosome-Targeting Chimera) technology at Stanford University, extending targeted protein degradation beyond the intracellular space to enable degradation of extracellular and membrane proteins through the endolysosomal pathway. Her LYTACs recruit the mannose-6-phosphate receptor or asialoglycoprotein receptor to direct target proteins for lysosomal degradation, opening new therapeutic possibilities for degrading cancer-relevant cell surface receptors and secreted proteins. This technology complements the PROTAC approach by addressing protein targets inaccessible to proteasomal degradation.
