Bert Vogelstein defined the multi-step model of colorectal carcinogenesis and identified p53 as the most commonly mutated gene in human cancer. His work established the genetic basis of cancer progression through sequential accumulation of oncogene activations and tumor suppressor losses. He pioneered liquid biopsy approaches using circulating tumor DNA for early cancer detection, transforming cancer diagnostics.
