Arlene Sharpe is a founding figure in the biology of PD-1 and co-inhibitory receptors governing T cell exhaustion. Her laboratory has mapped the epigenetic landscape of exhausted T cells, revealing stable chromatin changes that limit reinvigoration by checkpoint blockade. She demonstrated how TOX and NR4A transcription factors drive epigenetic programming of exhaustion. Her translational work has direct implications for overcoming IO resistance in cancer patients.
