Alan D'Andrea is a pioneer in the Fanconi anemia DNA repair pathway and its intersection with BRCA1/BRCA2-mediated homologous recombination. His laboratory discovered key components of the FA-BRCA pathway including FANCD2 ubiquitination and the nuclear complex that coordinates interstrand crosslink repair. His mechanistic work directly linked Fanconi anemia to BRCA breast and ovarian cancer predisposition, revealing the unified FA-BRCA DNA repair network. His translational research on PARP inhibitor sensitivity and BRCA reversion mutations has informed clinical biomarker development for PARP inhibitor therapy.
