👥Biography 个人简介

Miriam Merad at Icahn School of Medicine is a world leader in myeloid cell and dendritic cell biology. Her discoveries on tissue-resident macrophage ontogeny and the heterogeneity of tumor-infiltrating myeloid cells—enabled by single-cell technologies she helped pioneer—have reshaped understanding of tumor immune microenvironments and myeloid-targeted therapy.

🎓Key Contributions 主要贡献

Tissue-Resident Macrophage Ontogeny

Demonstrated that many tissue-resident macrophages, including microglia and Kupffer cells, are embryonically seeded and self-renew independently of bone marrow precursors, redefining macrophage classification relevant to tumor-associated macrophage subsets.

Tumor Dendritic Cell Heterogeneity

Used single-cell transcriptomics to map the diversity of conventional and plasmacytoid dendritic cell subsets within human tumors, identifying DC subsets critical for cross-presentation and anti-tumor T cell priming.

Myeloid Cell Targeting in Cancer

Developed strategies to repolarize immunosuppressive tumor-associated macrophages and boost dendritic cell function in vivo, enabling synergy with checkpoint blockade and demonstrating durable anti-tumor responses.

The dendritic cell lineage: ontogeny and function of dendritic cells and their precursors in steady state and the inflamed setting

Annual Review of Immunology (2013)

Definitive review of DC ontogeny, subset specification, and function across tissues that remains a standard reference for myeloid cell biologists studying tumor immunity.

Tissue-resident macrophages self-maintain locally throughout adult life with minimal contribution from circulating monocytes

Immunity (2013)

Landmark study establishing the embryonic origin and local self-renewal of tissue macrophages using parabiosis models, overturning the classic monocyte-replacement paradigm.
