👥Biography 个人简介

Matthew Krummel at UCSF is the co-discoverer of CTLA-4 as an inhibitory checkpoint and a pioneer of intravital two-photon microscopy for studying immune cell dynamics in tumors. His work bridging PD-1/CTLA-4 checkpoint biology with real-time imaging has fundamentally advanced understanding of T cell exhaustion and tumor immune exclusion.

🎓Key Contributions 主要贡献

Co-Discovery of CTLA-4 as Immune Checkpoint

Co-authored with James Allison the original 1995 Journal of Experimental Medicine study showing that CTLA-4 blockade with antibodies enhances T cell activation—the founding experiment behind ipilimumab and the checkpoint blockade revolution.

Intravital Two-Photon Imaging of Tumor Immunity

Developed and applied intravital two-photon microscopy to visualize T cell motility, contact dynamics, and functional states within live tumors, revealing how physical and metabolic barriers enforce immune exclusion and T cell dysfunction.

PD-1 and T Cell Dysfunction Mechanisms

Elucidated molecular mechanisms by which PD-1 signaling disrupts TCR signal transduction and cytoskeletal reorganization at the immune synapse, explaining how tumors exploit PD-L1 to disable infiltrating T cells locally.

Representative Works 代表性著作

CD28 and CTLA-4 have opposing effects on the response of T cells to stimulation

Journal of Experimental Medicine (1995)

Seminal paper demonstrating CTLA-4 as a negative regulator of T cell activation, providing the conceptual and experimental foundation for checkpoint immunotherapy.

Visualizing tumour immunity

Nature Reviews Immunology (2014)

Comprehensive review of intravital imaging approaches for studying immune cell behavior in tumors, establishing best practices and key findings from live tumor imaging studies.
