Nick Barker, working with Hans Clevers, co-discovered LGR5 as a marker of intestinal crypt base columnar stem cells, a landmark finding that identified the cell of origin for intestinal cancer and revolutionized understanding of intestinal homeostasis and cancer initiation. LGR5+ cells are now recognized as the cancer stem cell population in colorectal cancer maintained by Wnt signaling, and anti-LGR5 antibody-drug conjugates are in clinical trials. Barker has continued to characterize LGR5+ CSC populations across multiple epithelial cancers and developed LGR5-based reporter systems enabling real-time CSC tracking in organoid and in vivo models.
