Max Wicha is a pioneer of the cancer stem cell (CSC) field who identified cancer stem cells in human breast tumors and established the landmark functional assays and surface markers (CD44+/CD24- and ALDH activity) used worldwide to prospectively isolate breast CSCs. His laboratory demonstrated that breast CSCs exist in distinct epithelial and mesenchymal states interconnected by EMT plasticity, and that IL-6/JAK/STAT3 and Notch signaling maintain the CSC niche. Wicha has led clinical trials targeting CSC pathways including Notch, Hedgehog, and IL-6, advancing the CSC hypothesis from bench to therapeutic testing.
