Marcus Bosenberg is a melanoma pathologist and tumor immunologist who developed the BRAFV600E/PTEN knockout GEM (Tamoxifen-inducible genetically engineered mouse) model of melanoma that recapitulates the immunosuppressive tumor microenvironment of human melanoma with low TIL infiltration. This model was instrumental in demonstrating why immunotherapy can fail and in testing combination strategies to convert cold tumors into hot ones. Bosenberg has used this model to characterize how BRAF and PTEN mutations cooperate to create the immunosuppressive TME and has identified strategies including radiation and targeted therapy to overcome immune exclusion.
