👥Biography 个人简介
Lewis Cantley, PhD is a pioneering biochemist at Weill Cornell Medicine who discovered phosphoinositide 3-kinase (PI3K), one of the most frequently mutated enzymes in human cancer. His foundational work on PI3K/AKT/mTOR signaling transformed understanding of cancer metabolism, insulin resistance, and tumor growth, and catalyzed the development of PI3K inhibitors as cancer therapeutics.
🧪Research Fields 研究领域
🎓Key Contributions 主要贡献
PI3-Kinase Discovery
Discovered and characterized phosphoinositide 3-kinase (PI3K) and its activation by oncoproteins, establishing the PI3K/AKT/mTOR axis as a central driver of cancer cell growth and metabolism.
Fasting and Dietary Interventions
Demonstrated that insulin and IGF-1 signaling driven by dietary sugar promotes tumor growth and that fasting or low-sugar diets can enhance the efficacy of cancer therapy through metabolic mechanisms.
Representative Works 代表性著作
A phosphatidylinositol 3-kinase/Akt pathway promotes translocation of Mdm2 from the cytoplasm to the nucleus
Genes & Development (1998)
Established PI3K/Akt as a key survival pathway suppressing apoptosis in cancer cells.
Fasting-mimicking diet and hormone therapy induce breast cancer regression
Nature (2020)
Showed that dietary intervention mimicking fasting sensitizes breast tumors to hormone therapy via metabolic and endocrine mechanisms.
