Stephen Lessnick is a molecular biologist who has dedicated his laboratory to understanding the oncogenic mechanism of EWS-FLI1, the defining fusion transcription factor in Ewing sarcoma. His laboratory discovered that EWS-FLI1 activates gene expression through GGAA microsatellite-containing enhancers, revealing a unique mechanism by which oncogenic transcription factors hijack repetitive DNA elements to drive cancer. Lessnick has identified downstream EWS-FLI1 targets including NKX2.2 and ID2 as potential therapeutic vulnerabilities and developed platform approaches to assess EWS-FLI1 transcriptional activity as a pharmacodynamic biomarker in Ewing sarcoma clinical trials.
