James Herman is a pioneer in cancer epigenetics who discovered promoter hypermethylation as a universal mechanism of tumor suppressor gene silencing across cancer types. His identification of methylated CDKN2A/p16, DAPK, and MGMT as cancer biomarkers enabled DNA methylation-based detection approaches applicable to plasma, sputum, bronchial wash, and other clinical specimens. Herman co-developed the MSP (methylation-specific PCR) technique, which became the most widely used method for methylation biomarker analysis. His work underpins multiple methylation-based cancer detection assays now in clinical development.
