👥Biography 个人简介

Mikael Pittet is a leading authority on myeloid cell biology in the tumor microenvironment, with particular expertise in how tumor-associated neutrophils (TANs) and myeloid-derived suppressor cells (MDSCs) are educated by tumors to suppress anti-tumor immunity. His single-cell and spatial transcriptomic studies have resolved myeloid cell heterogeneity in human tumors, revealing TAN and MDSC subsets that correlate with immunotherapy resistance.

🎓Key Contributions 主要贡献

TAN Heterogeneity and Plasticity

Used single-cell RNA sequencing to define pro-tumor (N2) and anti-tumor (N1) tumor-associated neutrophil transcriptional states, identifying TGF-β as a key switch toward immunosuppressive phenotypes.

MDSC Mechanisms in Immunotherapy Resistance

Demonstrated that polymorphonuclear MDSCs expand in patients progressing on PD-1 blockade and suppress NK cell and T cell function via ROS and arginase-1.

Spatial Tumor Immunology

Pioneered spatial transcriptomic mapping of myeloid niches in human tumors, revealing how MDSC-rich exclusion zones physically prevent T cell infiltration.
