Janet Rowley was a pioneering cancer cytogeneticist who identified chromosomal translocations as causative events in leukemia, transforming understanding of the genetic basis of cancer. In 1972, she discovered that the Philadelphia chromosome in CML results from a specific reciprocal translocation t(9;22) rather than a chromosome deletion, and subsequently identified t(8;21) in AML and t(15;17) in acute promyelocytic leukemia. The t(15;17) discovery was the direct basis for retinoic acid differentiation therapy in APL, which cured a previously fatal leukemia. Rowley received the Presidential Medal of Freedom and National Medal of Science, and her discoveries directly enabled imatinib development by identifying BCR-ABL as the Philadelphia chromosome product.
